چکیده

The effect of hemicholinium-3 on the incorporation of [Me-%]choline into the phospholipids of the subcellular fractions of the canine caudate nucleus was studied. A correlation between the amount of hemicholinium-3 administered and increased choline incorporation into phospholipids was obtained. The analysis of the labeled phospholipids revealed that 93% of the radioactivity was in phosphatidylcholine and the remainder in lysophosphatidyl choline and sphingomyelin + choline plasmalogens. The specific radioactivity of phosphatidylcholine (countslmin per ,ug phosphorus) determined after its isolation by thin-layer chromatography, increased markedly as a result of the administration of hemicholinium. An unequal time-dependence in the labeling of the phosphatidylcholine in the subcellular fraction containing nerve endings, mitochondria and lysosomes, as opposed to the phosphatidylcholine of the microsomal fraction was also noted, particularly after the administration of hemicholinium and choline in a weight ratio of 500. Discontinuous density gradient centrifugation of the fraction containing the nerve endings, the mitochondria and the lysosomes showed that phosphatidylcholine formation was most affected by hemicholinium in the fraction containing the “heavy” nerve endings, followed by the fraction containing the mitochondria + lysosomes. It is suggested that hemicholinium acts by accelerating the intracellular rate of movement of choline from the water-soluble into the lipid pool of the caudate nucleus where, by virtue of its presencelQ, it subsequently also stimulates the incorporation of choline into membrane phosphatidylcholine.

Effects of cholesterol on the properties of equimolar mixtures of synthetic phosphatidylethanolamine and phosphatidylcholine. A 31P NMR and differential scanning calorimetry study.

1. The interaction of cholesterol with equimolar mixtures of 18 : l e /18 : I c phosphatidylethanolamine and 16 : 0/16 : 0 phosphatidylcholine has been investigated employing differential scanning calorimetry and 3,p NMR. At temperatures where lateral phase separation occurs cholesterol interacts preferentially with the (gel state) 1 6 : 0 / 1 6 : 0 phosphatidylcholine component , in agreement ...

Activation of intestinal mitochondrial phospholipase D by polyamines and monoamines.

Intestinal mitochondria have a phospholipase D (PLD) activity which was stimulated by polyamines and monoamines resulting in the formation of phosphatidic acid (PA) from endogenous phospholipids. When stimulated by polyamines, mitochondrial PLD utilized endogenous phosphatidylethanolamine (PE) as substrate whereas stimulated by monoamines, both PE and phosphatidylcholine (PC) were hydrolysed. S...

Control of choline oxidation in rat kidney mitochondria.

Choline is a quaternary amino cationic organic alcohol that is oxidized to betaine in liver and kidney mitochondria. Betaine acts as an intracellular organic osmolyte in the medulla of the kidney. Evidence is provided that kidney mitochondria have a choline transporter in their inner membrane. The transporter has a Km of 173+/-64 microM and a Vmax of 0.4+/-0.1 nmol/min/mg mitochondrial protein ...

Cytochrome c specifically induces non-bilayer structures in cardiolipin-containing model membranes.

(1) The effect of cytochrome c addition on the phospholipid structure of liposomes composed of cardiolipin, phosphatidylserine, phosphatidylglycerol, phosphatidylcholine or phosphatidylethanolamine in a pure form or in mixtures was investigated by 31P-NMR and freeze-fracture techniques. (2) Cytochrome c specifically induces the hexagonal Hii phase and possibly an inverted micellar structure of...

Polyunsaturated fatty acids stimulate phosphatidylcholine synthesis in PC12 cells.

The synthesis of phospholipids in mammalian cells is regulated by the availability of three critical precursor pools: those of choline, cytidine triphosphate and diacylglycerol. Diacylglycerols containing polyunsaturated fatty acids (PUFAs) apparently are preferentially utilized for phosphatide synthesis. PUFAs are known to play an important role in the development and function of mammalian bra...
